What USP Test Methods Are
USP test methods are standardized procedures published by the United States Pharmacopeia–National Formulary (USP–NF) that define how to test the identity, strength, purity, and quality of pharmaceutical ingredients, finished dosage forms, dietary supplements, and other regulated products. They are developed through expert committees and become enforceable reference points for manufacturers, regulators, and quality laboratories in the United States and beyond.

Unlike proprietary internal tests, USP methods are consensus-based, meaning they are reviewed by pharmacopeial experts from industry, academia, and government before they are adopted. The methods specify reagents, equipment, sample preparation, operating conditions, acceptance criteria, and calculations so that a result obtained in one laboratory can be meaningfully compared with a result from another.
Understanding which USP test method applies to a given material or product is a core part of quality assurance. Choosing the wrong method, or applying a method outside its validated scope, can lead to compliance findings, failed specifications, or inconsistent product performance.
Structure of USP–NF and How to Find the Right Method
The USP–NF is organized into monographs and general chapters. Monographs describe the tests and specifications for a specific substance or product, while general chapters cover analytical procedures and topics that apply across many entries.
Monographs
Each monograph typically includes sections for identification, purity, assay, and related substances. A monograph for a drug substance will list which USP test methods apply to that substance, along with the acceptance criteria that a batch must meet before it can be released. For example, a monograph might reference a specific HPLC method for assay and a separate dissolution apparatus configuration for a tablet dosage form.
General Chapters
General chapters, identified by a chapter number, provide the detailed procedural text. Examples include general chapters for water determination (Karl Fischer titration), residual solvents, particulate matter in injections, and dissolution testing. These chapters define the instrumentation, media, apparatus, and acceptance limits that the monograph then points to.
Major Categories of USP Test Methods
USP test methods span a wide range of analytical techniques. The most commonly used categories include:
- Identification tests — spectroscopic, chromatographic, or chemical procedures that confirm a material is what it is claimed to be.
- Assay methods — quantify the active ingredient, often by HPLC, UV-Vis spectrophotometry, or titration.
- Impurity and related substance tests — detect and quantify degradation products, process-related impurities, and residual solvents.
- Dissolution and release tests — measure how quickly a dosage form delivers the active ingredient under defined conditions.
- Microbiological methods — sterility testing, microbial limits, endotoxin testing, and preservative effectiveness.
- Physical tests — appearance, particulate matter, uniformity of dosage units, hardness, friability, and disintegration.
Method Validation and Suitability
USP test methods are not just procedures to follow; they also set expectations for method suitability. Before a method can be used for official testing, the laboratory typically performs a suitability check using a standard or spiked sample to confirm that the system, the analyst, and the conditions can produce reliable data.
Key parameters validated or verified include specificity, accuracy, precision, linearity, range, and detection or quantitation limits. For pharmacopeial methods, the user laboratory is responsible for verifying that the method, as written, is suitable for its intended use, unless the method has been previously validated and transferred with full documentation.
Compliance and Regulatory Use
USP test methods are referenced by the FDA and other regulatory agencies as part of drug approval, ANDA review, and post-market surveillance. A product that claims to meet a USP monograph must comply with every test and acceptance criterion listed in that monograph. During inspections, regulators may ask to see evidence that the methods are being performed correctly, that system suitability is documented, and that out-of-specification results have been investigated.
Because USP methods are incorporated into law through the Federal Food, Drug, and Cosmetic Act for drugs, following them is not optional for regulated products. For dietary supplements, USP methods serve as quality benchmarks, and the USP Dietary Supplement Verification Program relies on these procedures to confirm label accuracy and purity.
Staying Current with USP Revisions
USP revises its test methods regularly through its expert panels. Revisions may update acceptance criteria, introduce new analytical technologies, or harmonize methods with international pharmacopeias. Laboratories that depend on USP methods should monitor the USP–NF annual supplement, Notices, and proposed revisions to ensure their testing programs remain current and compliant.
